Title: 5f6-a bright channel

On Fender's schematic for the 5f6-a bright channel there is 100pf cap across the wiper. I dont have any 100pf so would a 250pf cap make much\difference?

It will make the 'bright' have a lower frequency response, so not quite so 'high' but you can definitely try it out and see if you like it.  If not order a 100pF and replace it.  It definitely won't hurt anything.

You can lookup the calculations out there for capacitor frequency and figure out for that amp the exact frequency 100pF vs 250pF and see the difference, but hearing it is probably best.

Two 250p wired in series is 125p, closer to what Leo picked.
